碘化丙啶PI 細(xì)胞檢測

說明：熒光染料PI（碘化丙啶）是一種可對DNA染色的細(xì)胞核染色試劑，常用于細(xì)胞凋亡檢測，英文全稱是Propidium Iodide。它是一種溴化乙啶的類似物，在嵌入雙鏈DNA后釋放紅色熒光。盡管PI不能通過活細(xì)胞膜，但卻能穿過破損的細(xì)胞膜而對核染色。PI經(jīng)常被用來與Calcein-AM或者FDA等熒光探針一起使用，能同時(shí)對活細(xì)胞和死細(xì)胞染色。PI-DNA復(fù)合物的激發(fā)和發(fā)射波長分別為535 nm和615 nm。

產(chǎn)品說明：
碘化丙啶（PI）是一種核酸熒光染料，它不能透過完整的細(xì)胞膜，但能夠透過死細(xì)胞和凋亡中晚期細(xì)胞的細(xì)胞膜而使細(xì)胞核染色。因此，PI 作為熒光探針常用于細(xì)胞凋亡（apoptosis）或細(xì)胞壞死（necrosis） 的檢測。在流式細(xì)胞分析中，PI 常與其他染料如 Calcein-AM、Hoechst 33258或 Hoechst 33342 聯(lián)合使用，來區(qū)分凋亡早晚期細(xì)胞以及死細(xì)胞。此外，PI 也常作多色熒光染色的復(fù)染劑使用。

產(chǎn)品用途：
用于細(xì)胞凋亡(apoptosis)或細(xì)胞壞死(necrosis)的檢測，常用于流式細(xì)胞儀分析；常被用來與 Calcein, AM、Hoechst 33258 或 Hoechst 33342 等一起使用，能同時(shí)對活細(xì)胞和死細(xì)胞染色。

使用方法: 可溶于水或 DMSO。染色過程如下：
1）用 PBS 或適當(dāng)?shù)木彌_液制備 10～50μM 的 PI 溶液。
2）將 1/10 培養(yǎng)基體積的 PI 溶液加入到細(xì)胞培養(yǎng)基中(也可以用 1/10 濃度的 PI 緩沖液代替培養(yǎng)基)。
3）在 37℃培養(yǎng)細(xì)胞 10～20 分鐘。
4）用 PBS 或合適的緩沖液洗滌細(xì)胞兩次。
5）用 535nm 激發(fā)波長，615nm 發(fā)射波長的濾光器的熒光顯微鏡觀察細(xì)胞。
